The potential involvement of G-quadruplex in biological functions, such as gene expression and regulation, has been increased as G-quadruplex has been shown to exist in vivo. While the formation of G-quadruplex in ssDNA has been well studied, we show, using single-molecule and chemical footprinting, the formation of human ILPR G-quadruplex in dsDNA under physiological pH and K+ ion concentration. We also present the evidence that the ILPR G-quadruplex is mechanically stable with an unfolding force of 24 pN. Such a stable tetraplex structure in ILPR dsDNA is potential to control the transcription regulation of insulin gene
