Abstract

In bioethanol production, hydrolysis step is responsible for sugar formation which further affects ethanol yield. The aim of this study is to compare the effect of pretreatment conditions and enzymatic hydrolysis conditions on rice straw digestibility. In this experimental study, the effect of pretreatment conditions, including alkali type and dose, on hydrolysis efficiency was tested. Further, the effects of enzymatic hydrolysis parameters including enzyme type, enzyme dose and substrate load on hydrolysis efficiency were investigated. Rice straw samples pretreated using NaOH, Ca(OH)2 or mixture of them were subjected to enzymatic hydrolysis at constant temperature and time. The highest reducing sugars value was obtained for 1% NaOH pretreated sample. Samples of rice straw treated with 1% NaOH were subjected to hydrolysis using two commercially available cellulase enzymes with doses of 2-8 ml/100g of substrate at 50°C for 24h. The study conducted the significant role of pretreatment, enzyme characteristics, loading and S/L ratio on both sugar concentration and saccharification efficiency.
